    Research shows that mathematics education in the U.S. is struggling to keep up with the rapid growth in Science, Technology, Engineering, and Math career opportunities, especially in secondary education. This project outlines several viewpoints on the best practices in college preparatory mathematics courses. After an extensive amount of research and analysis, examples of teaching modules have been created to enhance a high school student’s experience, while better preparing him/her for college level courses. Each topic is outlined using high school and college level applications. Data summarizing classroom experiments performed on high school students justify the necessity and highlight the success of a new way of thinking in mathematics

    Differences between murine arylamine N-acetyltransferase type 1 and human arylamine N-acetyltransferase type 2 defined by substrate specificity and inhibitor binding

    Background: The mouse has three arylamine N-acetyltransferase genes, (MOUSE)Nat1, (MOUSE)Nat2 and (MOUSE)Nat3. These are believed to correspond to (HUMAN)NAT1, (HUMAN)NAT2 and NATP in humans. (MOUSE)Nat3 encodes an enzyme with poor activity and human NATP is a pseudogene. (MOUSE)Nat2 is orthologous to (HUMAN)NAT1 and their corresponding proteins are functionally similar, but the relationship between (MOUSE)Nat1 and (HUMAN)NAT2 is less clear-cut. Methods: To determine whether the (MOUSE)NAT1 and (HUMAN)NAT2 enzymes are functionally equivalent, we expressed and purified (MOUSE)NAT1*1 and analysed its substrate specificity using a panel of arylamines and hydrazines. To understand how specific residues contribute to substrate selectivity, three site-directed mutants of (MOUSE)NAT2*1 were prepared: these were (MOUSE)NAT2_F125S, (MOUSE)NAT2_R127G and (MOUSE)NAT2_R127L. All three exhibited diminished activity towards “(MOUSE)NAT2-specific” arylamines but were more active against hydrazines than (MOUSE)NAT1*1. The inhibitory and colorimetric properties of a selective naphthoquinone inhibitor of (HUMAN)NAT1 and (MOUSE)NAT2 were investigated. Results: Comparing (MOUSE)NAT1*1 with other mammalian NAT enzymes demonstrated that the substrate profiles of (MOUSE)NAT1 and (HUMAN)NAT2 are less similar than previously believed. Three key residues (F125, R127 and Y129) in (HUMAN)NAT1*4 and (MOUSE)NAT2*1 were required for enzyme inhibition and the associated colour change on naphthoquinone binding. In silico modelling of selective ligands into the appropriate NAT active sites further implicated these residues in substrate and inhibitor specificity in mouse and human NAT isoenzymes. Conclusions: Three non-catalytic residues within (HUMAN)NAT1*4 (F125, R127 and Y129) contribute both to substrate recognition and inhibitor binding by participating in distinctive intermolecular interactions and maintaining the steric conformation of the catalytic pocket. These active site residues contribute to the definition of substrate and inhibitor selectivity, an understanding of which is essential for facilitating the design of second generation (HUMAN)NAT1-selective inhibitors for diagnostic, prognostic and therapeutic purposes. In particular, since the expression of (HUMAN)NAT1 is related to the development and progression of oestrogen-receptor-positive breast cancer, these structure-based tools will facilitate the ongoing design of candidate compounds for use in (HUMAN)NAT1-positive breast tumours. </p

    National Register Testing At Sites 41BP585, 41BP594, And 41BP595 Three Oaks Mine, Bastrop County, Texas

    Between October 2012 and July 2013, Atkins conducted National Register of Historic Places (NRHP) eligibility testing at historic sites 41BP585 and 41BP594 and prehistoric site 41BP595, located within the Three Oaks Mine in Bastrop County, Texas, which is owned and operated by Luminant. Impacts to all three sites are anticipated as a result of planned mine development. This work was conducted under the direction of Principal Investigator David L. Sherman. This report of investigations was written at Atkins and is being finalized by Blanton & Associates, with David L. Sherman remaining as the Principal Investigator. This work demonstrated that significant archeological deposits that may contribute to the overall NRHP eligibility statuses of the two historic sites are absent at both sites. Standing architecture at 41BP594, however, has previously been determined to be eligible for listing on the NRHP (Martin 2001). Archival research conducted as part of the current investigation into the histories of the historic sites remains inconclusive with respect to the identity of their 1870s and earlier occupants. Testing at prehistoric site 41BP595 indicated it resulted from multiple occupational episodes during the period from the late Paleoindian to the Late Prehistoric. Shovel testing and mechanical trenching revealed the presence of an expansive buried anthrogenic A soil horizon, or midden, replete with preserved subsistence remains. Mechanical trenching also exposed a variety of burned rock cooking facilities partially surrounding the midden area. Radiocarbon assays of burned nut shells recovered from feature contexts, along with the assemblage of diagnostic lithic artifacts, suggest the site was most intensively occupied from the Late Archaic to the early Late Prehistoric. A suite of special studies was conducted on burned rock samples recovered from four of the better-preserved burned rock features. These studies, which include residue, starch, and phytolith analysis, suggest that the burned rock features were used in part to process tubers/roots and grass seeds for subsistence. Macrobotanical analysis of flotation samples recovered from feature contexts identified spent fuel remains including oak and hickory wood and subsistence remains including oak, hickory, black walnut, and acorn burned nut shells. A small amount of burned bulb, possibly representing wild onion, was also recovered through flotation. These findings suggest that significant archeological deposits important to understanding the Late Archaic to early Late Prehistoric period have been preserved at 41BP595

    Incisional hernia following colorectal cancer surgery according to suture technique: Hughes Abdominal Repair Randomized Trial (HART).

    BACKGROUND: Incisional hernias cause morbidity and may require further surgery. HART (Hughes Abdominal Repair Trial) assessed the effect of an alternative suture method on the incidence of incisional hernia following colorectal cancer surgery. METHODS: A pragmatic multicentre single-blind RCT allocated patients undergoing midline incision for colorectal cancer to either Hughes closure (double far-near-near-far sutures of 1 nylon suture at 2-cm intervals along the fascia combined with conventional mass closure) or the surgeon's standard closure. The primary outcome was the incidence of incisional hernia at 1 year assessed by clinical examination. An intention-to-treat analysis was performed. RESULTS: Between August 2014 and February 2018, 802 patients were randomized to either Hughes closure (401) or the standard mass closure group (401). At 1 year after surgery, 672 patients (83.7 per cent) were included in the primary outcome analysis; 50 of 339 patients (14.8 per cent) in the Hughes group and 57 of 333 (17.1 per cent) in the standard closure group had incisional hernia (OR 0.84, 95 per cent c.i. 0.55 to 1.27; P = 0.402). At 2 years, 78 patients (28.7 per cent) in the Hughes repair group and 84 (31.8 per cent) in the standard closure group had incisional hernia (OR 0.86, 0.59 to 1.25; P = 0.429). Adverse events were similar in the two groups, apart from the rate of surgical-site infection, which was higher in the Hughes group (13.2 versus 7.7 per cent; OR 1.82, 1.14 to 2.91; P = 0.011). CONCLUSION: The incidence of incisional hernia after colorectal cancer surgery is high. There was no statistical difference in incidence between Hughes closure and mass closure at 1 or 2 years.     West Coast Communities and Catch Shares: The Early Years of Social Change

    <p>The Pacific Coast Groundfish Trawl Fishery transitioned to a catch shares program in January 2011. The Pacific Coast Groundfish Fishery Social Study was designed to measure associated social changes and impacts on individuals and communities. Selected survey and interview data from the baseline data collection in 2010 and the first supplemental data collection effort in 2012 are aggregated at the community level and analyzed for initial signs of social change.</p> <p>Communities are sorted into top-, mid-, and low-tier communities based on the percentage of quota share (QS) permit owners that live in each community. A higher number of QS permit owners in a place is expected to result in relatively greater benefits to those communities. Questions analyzed include percent of income from fishing, multiple jobs worked, job stability, job satisfaction, standard of living, and how individuals were personally affected. Significant results for TOP-tier communities include increases in working multiple jobs, which may be negative or positive depending on the respondent, and improvements in job satisfaction. LOW-tier communities indicate significant improvements in standard of living. MID-tier communities appear to be in the middle, with no significant changes. Interview data indicate variation between owners, where some can fish their allocations and others need to lease more to fish.</p

    <div><p>Human arylamine <i>N</i>-acetyltransferase 1 (hNAT1) has become an attractive potential biomarker for estrogen-receptor-positive breast cancers. We describe here the mechanism of action of a selective non-covalent colorimetric biosensor for the recognition of hNAT1 and its murine homologue, mNat2, over their respective isoenzymes, leading to new opportunities in diagnosis. On interaction with the enzyme, the naphthoquinone probe undergoes an instantaneous and striking visible color change from red to blue. Spectroscopic, chemical, molecular modelling and biochemical studies reported here show that the color change is mediated by selective recognition between the conjugate base of the sulfonamide group within the probe and the conjugate acid of the arginine residue within the active site of both hNAT1 and mNat2. This represents a new mechanism for selective biomarker sensing and may be exploited as a general approach to the specific detection of biomarkers in disease.</p></div
